Determination of rotenone in Derris elliptica by reversed-phase high performance liquid chromatography

Abstract: Objective:To establish a reversed-phase high performance liquid chromatography method for rotenone in Derris elliptica.Methods:The determination was performed on ZORBAX Eclipse XDB-C18 (5 μm,4.6×150 mm)column. The mobile phase consisted of acetonitrile and 0.1% phosphate acid solution(4852,v/v,pH 2.6)with flow rate at 1.0 mL/min and determination wavelength at 299 nm under 40.0 ℃.Results:Under the conditions described, the calibration curve of rotenone was linear over the concentration range of 30 to 50 mg/L. The regression equation was: A=20.731 1C-0.361 6, r=0.999 8(n=5,P<0.01). The average recovery for rotenone was 100.74%. The mean relative standard deviation(RSD) was 0.86%.Conclusion:The method is simple and accurate, and suitable for the determination of rotenone in Derris elliptica.
