Determination of chlorogenic acid content in wujin capsule by RP-HPLC

  • Objective To establish a RP-HPLC method for determination of chlorogenic acid in wujing capsule. Methods Agilent extend-C18 (4.6 mm×250 mm,5 μm)column was used. The mobile phase was acetonitrile: 0.1%formic acid (8:92). The flow rate was 1.0 ml/min, and the temperature was at 35 ℃. The detection wavelength was 327 nm. Determination of chlorogenic acid in wujin capsule of 5 batches of preparations. Results The calibration curve was at the range of 2.04~102 μg/ml(r=0.999 9), and the average recovery was 104.68 %,and RSD was 1.43%(n=6).5 batches of chlorogenic acid contents in wujin capsule were between 55.08~83.31 μg/g. Conclusion The method was sensitive and accurate, and could be used for the quality control of wujin capsule.
