HPLC法测定山楂麦曲颗粒中金丝桃苷的含量

摘要: 目的:建立测定山楂麦曲颗粒中金丝桃苷的方法。方法:采用HPLC法,色谱柱:Agilent ZORBAX SB-C18柱(4.6 mm×250 mm,5μm),流动相:甲醇-四氢呋喃-0.5%磷酸溶液(0.9:17.6:81.5),流速:1.0 mL/m in,检测波长:360 nm,柱温:室温;进样量:20μL。结果:金丝桃苷在0.018 624~0.465 6μg范围内呈良好的线性关系(r=0.999 9);平均回收率为96.097%,RSD为0.82%(n=5)。10批山楂麦曲颗粒中金丝桃苷的含量测定结果在每袋含49~65μg。结论:该方法操作简单、重现性好、可用于山楂麦曲颗粒的质量控制。Abstract: Objective :To establish a method for determination of hyperoside in Shanzha Maiqu granule. Methods :HPLC was adopted with Agilent ZORBAX SB-C18 column(4.6 mm×250 mm,5 μm),methanol-tetrahydrofuran-0.5% H3PO4(0.917.681.5) as mobile phase at flow rate of 1.0 mL/min and 360 nm as detecting wavelength.Column temperature was room temperature and sample size was 20 μL. Results :The linear range of hyperoside was 0.018 624~0.465 6 μg(r=0.999 9).The average recovery rate of hyperoside was 96.09% and RSD was 0.82%(n=5).The contents of hyperoside in ten batches of Shanzha Maiqu granule were deteminted between 49~65 μg. Conclusion :This method is simple,accurate and reproducible,suitable for quality control of the drug.
