Abstract:
Objective In order to understand the regulation of hypoxia on the vascular endothelial growth factor (VEGF) permeability action, particularly in relation to the risk of development of atherosclerosis, the effect of recombinant human VEGF on the oxidation low density lipoprotein (OxLDL) permeability of bovine aortic endothelial cells (BAEC) was observed under the hypoxia condition in vitro.Methods Under normal or hypoxia condition, the confluence BAEC monolayers were cultured with normal medium and with medium containing VEGF at various concentrations and for various time periods. The Iodine labeled low density lipoprotein flux across the monolayers was then performed, and the radioactivity was measured by SN 695 automatic liquid scintillation counter. Results Addition of VEGF to the BAEC monolayers significantly increased the permeability of the monolayer to [125I]OxLDL (P<0.01). The permeability increasing activity of VEGF on the BAEC monolayers was concentration dependent. Hypoxia injury on endothelial cells increased VEGF induced hyperpermeability. Conclusion VEGF plays a role in the formation and development of atherosclerosis.

Abstract:
Objective To observe the effect of a novel liposome-prednisolone on puromycin aminonucleoside (PAN) nephropathy model in rats. Methods 160~180g BW male Wistar rats were used in this experiment. The rats were given a single injection of PAN at the dose of 4mg/100g BW through right jugular vein to establish the nephrotic syndrome (NS) model. The free prednisolone (PRED) treated group were given (i.p.) the free prednisolone at the dose of 2.0mg/100g BW and the liposome prednisolone (Lipo-PRED) treated group were given (i.v.) the liposome prednisolone at the dose of 1.5mg/100g BW. Urinary protein excretion examined with the biuret method and the monocyte/macrophage, CD4+ T cells and CD8+T cells were analyzed by immunohistochemistry. Results The proteinuria of the Lipo-PRED treated group reduced; the infiltration of monocyte/macrophages, CD4+T cells and CD8+T cells in the glomeruli and interstitium also remarkably weakened. Conclusion The liposome prednisolone can remarkably reduce the proteinuria excretion and leukocyte infiltration both in the glomeruli and tubulointerstitium in the PAN model.

Abstract:
Objective To study the formation of transdermal delivery system of lidocaine with lecithin microemulsions as delivery system.Methods Area of water in oil microemulsions was investigated through pseudo ternary phase diagrams. Viscosity of microemulsions were measured to select the the right formulation using orthogonal test method. The content of hydrochloride lidocaine in microemulsions was measured by spectrophotometric. Results Nature of cosurfactant and K m showed predominant influence on the area of microemulsions, area of microemulsions in propanol and iso propanol system were larger. The area of microemulsions was larger in low purity lecithin system. Cosurfactant showed predominant influence on the viscosity of the microemulsion. Content of lidocaine was measured at 262nm, the standard curve was linear in the range of 0.02~0.5mg/mL, the average recovery was (100.17±0.16)%, the within day and between day precision of determination of lidocaine was 0.0137±0.0121, 0.0119±0.0115. Conclusion Choosing mixed surfactant will help the formation of microemulsion. Nature and potent of alcohol have very important effect on selection of the formulation of lidocaine transdermal microemulsions.

Abstract:
Objective To evaluate the clinical therapeutic outcomes of Yigankang in treatment chronic hepatitis B patients.Methods 31 patients with HBeAg(+),HBV-DNA(+)were treated by routine therapy with Yigankang for 3 months,and 25 patients as control were investigated meantime.Results The serum HBeAg negative conversion rate was 35.5% and the serum HBV-DNA negative was 45.1% after treatment.The serum HBeAg and HBeAb conversion rate was 29%.Conclusion Yigankang can improve the liver function of chronic hepatitis B,and it can eliminate HBV.

Abstract:
Objective To offer reference for selecting effective antibacterial drugs in the clinic treatment through dynamic monitoring the pathogenic bacterium of 200 aged patients with respiratory tract infection(about sixty years or older) and drug susceptibility test.Methods The sputum from patients was collected and monitored .The pathogenic bacteria in the sputum separated and identified.The drug susceptibility test was used by K-B method and the results were judged according to NCCLS.Results The major pathogenic bacteria in old respiratory tract infection were gram negative bacteria,the infection rate was about 81.2%(125strains).The infection rate of gram-positive bacteria was about 18.8%(29 strains).The fungus infection rate was higher about 48.8%(147 strains).The bacteria variance is very quick.Conclusion A dynamic monitoring and drug susceptibility to pathogenic bacterium play an important role in the clinic treatment.

Objective To improve the dissolution rate of nifedipin tablets in vitro.Methods The effects of particle size, diluents and disintegrator on dissolution of nifedipine tablets in vitro were studied, compare dissolution curve method was used to optimize the formulation. Results The nifedipine tablets which were prepared with optimized prescription had the excellent quality of dissolution and have achieved the quality of Parmacopeia of Peoples Republic of China (2000). Conclusion The formulation is reasonable, it is suitable for manufacturing industrally.

Objective To establish a quantitative analysis method for camphor in compound menthol nasal drops.Methods The camphor was determined directly at (292±1)nm by ultraviolet spectrophotometry.Results:The calibration curve was linear in the range from 1.62~3.24mg/mL with r=0.9992,the average recovery and relative standard deviation were 100.06% and 1.29% respectively. Conclusion The method is simple,rapid and accurate.

Objective To establish a method to determinate scopolamine hydrobromide injection by ion pair RP-HPLC.Methods The mobile phase of methanol-20mmol/L sodium heptanesulfonate solution acetic acid (50:50:2) and ZORBAX SB-C18 column (4.6×250mm,5μm)was used,the flow rate was 0.9mL/min and the detect wavelength was 225nm.Results The linear range of scopolaminc hydrobromide was 0.1~0.5mg/mL(r=0.9999). The average recovery was 100.15%,RSD was 1.58%.Conclusion The method was simple,rapid and accurate. It can be used to control the quality of scopolamine hydrobromide injection.

Objective To establish a HPLC method for determination of acetaminophen in compound paracetmol and zinc gluconate tablets. Methods Alltech-C18 (4.6×250mm,5μm)column was used and the mobile phase consisted of methanol and water (20:80). It was detected at 246nm. Results Linearity was obtained in the concentration rage of 4~60μg/mL, A=-13496.960 9+78302.6211C(r=0.9999).The average recovery was 99.83%,RSD=0.93%(n=6). Conclusion The method provide an accurate and sensitive way for the determination of acetaminophen in compound paracetmol and zinc gluconate tablets by HPLC.

Objective To establish a method of HPLC for determination of forsythin in Kangbingdu oral liquid.Methods A LUNA C18 (2) column was used. The mobile phase was acetonitrile-water(25:75).The flow rate was 0.9mL/min.The detection wavelength was set as 277nm. Results The linear range of forsythin was 0.46~2.3μg,and the average recovery rate was 100.06%,RSD=1.0%. Conclusion The method is simple, reliable, and accurate and can be applied to the quality control of preparation.
