New progress of protein crystallization and drug design

Abstract:
Protein was the foundation of all life, and its function was closely related to its three-dimensional structure. The structure information of proteins was crucial for the determination of drug targets and helped scientists to discover new drug targets according to the structure homology between the protein targets and drugs. Therefore, the availability of protein crystal, which was the basis for its structure information and the drug design, had become an important field in life science. The latest research progresses of protein crystallization technology and its application in the drug design were summarized in this review.

Abstract:
Micro-needles transdermal drug delivery system became the hot spot in the field of transdermal drug delivery system, which had the double advantages in both injection drug delivery and transdermal drug delivery. It also had the merits of fast absorption, continence and painless utility in clinical. Research indicated both transdermal speed and dose of drugs were highly increased when micro-needles transdermal drug delivery system was applied, which especially showed outstanding effects and promise in the preparation of drugs composed of protein, polypeptide, DNA and RNA, etc. Latest developments of micro-needles transdermal drug delivery system were reviewed in this paper. With the development of the industrial arts of the micro-needle, drug delivery technology and application research, micro-needles transdermal drug delivery system would be extensively employed in clinical field.
Advance in function of the eukaryotic elongation factor eEF1A

Abstract:
Eukaryotic elongation factors(eEFs)was a kind of important molecules that assist in elongating the polypeptide chain in the protein biosynthesis of the organisms. It had two subtypes in eukaryotic organism:eEF1 and eEF2. One subtype of eEF1:eEF1A played the main functions in elongating the peptide chain, which carried the aminoacyl-tRNA (aa-tRNA)on the A site of the ribosome,and many studies indicated that eEF1A played another roles range from cell apoptosis,tumorigenesis,signal transduction to the regulation of the cardiovascular system, and these functions were not related to the protein synthesis. All of this demonstrated eEF1A was a multifunction protein,which could be a potential target of the disease prevention and cure. The advance in function of the eukaryotic elongation factor eEF1A in recently years were introduced in this article.
Advances in active ingredients and action mechanism of TCM in treatment of hepatic fibrosis

Abstract:
Hepatic fibrosis was the common feature of pathology in almost all patients with chronic liver injury, which led to cirrhosis gradually and liver cancer ultimately. The anti-hepatic fibrosis active ingredients from TCM, including flavones, alkaloids, terpenes and phenols compounds were reviewed with the action mechanism in this paper, which could provide some references for the further study about the anti-hepatic fibrosis of TCM.

Abstract:
Objective To introduce advances of anti-HBV bioactive substances from traditional Chinese medicine. Methods Anti-HBV constituents from traditional Chinese medicine research were reviewed, which were focused on the saponins, alkaloids, flavones and polysaccharides. Results and Conclusions Traditional Chinese medicine had been demonstrated as the potent anti-HBV bioactive substances, which was the important approach to the treatment of HBV.

Abstract:
Objective To study the chemical constituents of the South China Sea gorgonian Muriceopsis flavida. Methods The compounds were isolated and purified by repeated column chromatographies including normal-phase silica gel, Sephadex LH-20 and semi-preparative RP-HPLC. The structures were elucidated by the physicochemical evidences and spectra data. The in vitro bio-activities of the compounds were assessed by an agar diffusion test. Results Two sesquiterpenes were isolated and indentified as menverins C (1) and D (2). The two compounds showed different levels of antimicrobial activity in bioassay in vitro. In particular, compounds 1 and 2 displayed considerable growth inhibition toward bacterium Bacillus migaterium and the alga Chlorella fusca, respectively. Conclusions This was the first report of menverins C (1) and D (2) from the genus Muriceopsis. The antimicrobial activity of both compounds suggested a possible ecological role of these metabolites in the chemical defensive strategy of the gorgonian.
Protective effect of α-lipoic acid on noise induced hearing loss in guinea pigs

Abstract:
Objective To observe the protection of free radical scavenger: α-lipoic acid on the explosive hearing loss in rats. Methods 24 pure white male guinea pigs weighted between 250~300 g, which were randomly divided into α-lipoic acid low, medium, high group(n=3×6), Noise controlled group(n=6). Each group of animals was exposed to octave band noise (4 kHz center frequency) 115 dB SPL 4 h. α-Lipoic acid group were administered α-lipoic acid 30 mg/(kg·d), 60 mg/(kg·d), 120 mg/(kg·d) for 10 days (2 days before the exposure, 7 days after); Noise controlled group animals were given normal saline orally. After exposure, auditory brainstem response (ABRs) was tested in animals in each group at different times; hair cell loss and damage were examined through histological means. Results After exposure of 3 days, lower/middle/ high dose group had significant differences with noise control group at frequencies 8, 16 kHz; low and high dose group had significant differences with noise control group at frequencies 24, 32 kHz; 7 days after exposure, middle/ high dose group had significant differences with noise control group at frequencies of 8, 16, 24 kHz), low and high dose group had significant differences with noise control group at frequencies 32 kHz; after 10 days, high dose group had significant differences with noise control group at each frequencies. In the histological examination, animals of three α-lipoic acid dose group were found little inside and outside hair cell loss or damage. Noise controlled animals showed outer hair cells in the ear edema/vacuolar degeneration and other diseases. Conclusions Low, middle, high doses of a-lipoic acid on the explosive hearing loss had a preventive effect. High dose α-lipoic acid could take a good prevention and treatment in noise-induced hearing loss.

Abstract:
Objective To establish a method for determination of bacterial endotoxin in human coagulation factor Ⅷ by kinetic turbidimetric limulus test. Methods Based on interference experiment, the endotoxin was added in the samples, whose recovery should be within the range 50%~200%. Results The interference from limulus test was eliminated by diluting the Human coagulation factor Ⅷ (1→100 solution). Conclusions Turbidimetric-kinetic method was feasible, and had the same result with the rabbit-pyretogen test.

Abstract:
Objective To establish, compare and evaluate the classification models of antibiotic prophylactic use for cesarean section patients for the targeted intervention in future. Method PASW® Modeler 13 was applied to establish classification models and to get the influential variables (clinical factors) in antibiotic prophylactic use. Results With the data of 787 cases, the classification models were established, in which, Bayesian networks, logistic regression and CHAID were better. In 21 clinical factors, blood loss was the most influential variable. Conclusion The data mining technique was able to quickly create models reflecting the use of prophylactic antibiotics use for cesarean section, which would provide a new analysis tool for drug use survey.
Analysis of volatile constituents in Caulis Sinomenii by GC-MS

Abstract:
Objective To establish the determination method for chemical constituents of the volatile oil in Sinornenium actum(Thunb.)Rehd.et Wils by gas chromatography-mass spectrometry(GC-MS). Method The volatile oil was extracted from Sinornenium actum(Thunb.)Rehd.et Wils by steam distillation.The constituents of volatile oil were identified by GC-MS technology,which were identified through database index. Results The results showed that after comparing of the gained mass spectrum with the standard, 63 kinds of chemical constituents in all were identified, the major constituents were the fatty acids, sterols, alkenes and a few kind of aldehyde. The most contents were octadecylic acid and hexadecanoic acid, whose relative percentages were 33.78% and 23.23% respectively. Conclusion The experiment improved the basic components study on Caulis Sinomenii, which could provide more reference quality control of Caulis Sinomenii.

Abstract:
Objective To determine the sulfate content in sulfated fucans (FPS) by Barium sulfate turbidity. Methods With spectrophotometer at 360 nm, the sulfate content was determined by Barium Sulfate Turbidity. Results There was a good linearity (r2=0.990 9) when the sulfate content was within the range of 60~120 μg. Sulfate content of FPS was (20.52±0.57)% under the determination method.Methodology met the measurement requirement. Conclusion The method was proved to be reliable, quick, sensitive and applicable.

Abstract:
Objective To investigate effects of total aglycone of centella asiatica on depressed rats and study plasma levels of adrenocorticotropic hormone (ACTH) and cortisol (CORT). Methods The chronic mild stress depressed rats were produced by long-term unpredicted mild stress. The behavior of rats was detected by open field test. Objective index of anhedonia were sucrose water consumption and preference percentage of sucrose water in liquid consumption test. The levels of plasma ACTH and CORT were measured by radioimmunoassay. Results The depressed rats experienced less squares crossed, rears in the open-field test. Sucrose consumption in the sucrose preference test decreased significantly. The concentration of plasma ACTH and CORT significantly increased in stress-model rats compared to the control rats. The behavioral disorders and the levels of plasma ACTH and CORT of rats were improved by total aglycone of centella asiatica significantly. Conclusion Total aglycone of centella asiatica had anti-depressant effects, and which mechanism might be related to the improvement of hypothalamus-pituitary gland-adrenal glands axis function.

Abstract:
Objective To observe the clinical efficacy of Chanhuning and ganciclovir in treatment of children with infections mononucleosis. Method 76 patients were randomly divided into the treatment group (38 cases) and the control group (38 cases), the treatment group was treated with Chuanhuning and ganciclovir, the control group was only treated with ganciclovir. The clinical efficacy of each group was analyzed. Results The extinction times of clinical symptoms and signs subside, atypical lymphocytes and fever in treatment group were obviously less than those in control group, the clinical efficacy of the treatment group was significantly superior to the control group (P<0.05). Conclusion Chuanhuning combined with ganciclovir to treat children with infections mononucleosis was safe and effective, which could effectively shorten the cooling time and be worthy of clinical application.
Analysis on rationality of antibiotics choice after blood culture and drug sensitivity tests

Abstract:
Objective To analysis the distribution and drug resistance of pathogens isolated from blood cultures in hospitalized patients in order to guide the rational application of antibiotics agents. Methods 230 cases of blood culture results in hospitalized patients during 2009 October to 2010 October were retrospectively analyzed. Results 63 strains of pathogenic bacteria were checked out. The detection rate gram-negative bacteria, gram-positive bacteria was 80.9%, 17.51%, 1.59% respectively. Main pathogenic bacteria were Escherichia coli, Klebsiella spp, Enterobacter, Pseudomonas aeruginosa, with detection rate of 20.63%, 12.7%, 9.52%, 9.52% respectively. The detection rate of E.faecalis, Staphylococcus epidermidis, and E.faecium were 9.52%, 1.59%, 1.59%. Detection rate of Escherichia coli and Klebsiella spp induced ESBL was 6.34% and 1.59%. The susceptibility of gram-negative bacteria to carbapenem, amikacin, piperacillin/tazobatam were above 84%. Conclusion Glycopeptide antibiotics such as vancomycin were the first choice to cure serious blood infection caused by gram-positive bacterial. Amikacin, β-lactam/β-actamase inhibitors compound, carbapenem were the first choice to cure serious blood infection caused by gram-negative bacterial.
Meta-analysis of gemcitabine versus vinorelbine combined with cisplatin in treatment of advanced non-small cell lung cancer

Abstract:
Objective To compare and evaluate the effectiveness and security of gemcitabine versus vinorelbine combined with cisplatin in treatment of advanced non-small cell lung cancer by Meta-analysis. Methods PubMed database and CHKD database were searched, included in randomized controlled trials. Special software Review Manager Version 4.2.2 was used to systematic reviews. Results Seven studies of English journals involving 1 561 patients and ten studies of Chinese journals involving 864 patients were included. There was no difference in the overall response from gemcitabine versus vinorelbine in combination with cisplatin by Meta-analysis of English and Chinese journals. But the project of gemcitabine+cisplatin was superior to vinorelbine+cisplatin in 1-year survival rate of English journals and there was no difference in 1-year survival rate by Meta-analysis of Chinese journals. Both English and Chinese journals showed the same reports of toxicity. The incidence rate of neutropenia was higher in vinorelbine+cisplatin, and the thrombocytopenia was higher in gemcitabine + cisplatin, nausea and vomiting and nervous system response were no significant difference between two groups. Conclusions The effectiveness of gemcitabine versus vinorelbine combined with cisplatin of advanced non-small cell lung cancer was different. Improving the quality of RCT in Chinese journals was very important.
Meta analysis on cinepazide maleate in treatment of diabetic peripheral neuropath

Abstract:
Objective To assess the efficacy and safety of Cinepazide Maleate(CPZD) in the treatment of diabetic peripheral neuropath(DPN),with evidence-based medicine methods. Methods By retrieving related database,such as Medline(1977~2011.11), CNKI(1994~2011.11), WANFANG(1989~2011.11), VIP(1989~2011.11), the randomized controlled trials on DPN treatment by CPZD were included and analyzed according to the inclusion criteria.Meta analysis was used in this study. Results There were 982 cases of 11 trials were included in the analysis, the methodological quality of all studies included were rated as C-class. Meta-analysis showed that: 1Efficient: CPZD group was significantly higher than control group[OR and 95% CI=4.33 (2.45,7.66)]2Improvement of neurological function: The combined effect size of common peroneal nerve(CPN) SNCV and MNCV, median nerve(MN) SNCV and MNCV were as follows, CPN-SNCV:WMD=2.52,95%CI(1.92,3.13);CPN-MNCV:WMD=3.07,95%CI(1.59,4.55);MN-MNCV: WMD=3.97,95%CI(1.87,6.07);MN-SNCV:WMD=2.94,95%CI(1.43,4.46), the above-mentioned value were significantly higher than control group. 3Adverse reactions were less, mainly showed mild headache, dizziness, gastrointestinal discomfort. Conclusions Cinepazide Maleate might have positive effect on DPN. However,the evidence was not strong enough due to the general low methodological quality.A reliable conclusion about the effects of CPZD on DPN could not be summarized at the moment. Further large high-quality randomized controlled trials should be done.

Abstract:
Objective To explore practical measures to reduce the pharmacies dispensing errors, and improve the quality of drug deployment work. Methods The dispensing errors recorded in the Drug deployment errors registration from January 2009 to December 2011, were collected and analyzed. Results The total quantity of deployment errors was 101 in three years, which had 28 cases of number error, 20 cases of species errors, 12 cases of packaging similar errors, 11 cases of dose/formulation errors, 11 cases of manufacturer errors, 6 cases of wrong priced, 5 cases of drug repercussion, 4 cases,of prescription errors 2 cases of dispensing medicine to wrong department, 2 cases of taking medicine from nearly wrong place. Conclusions A drug deployment standardized system was gradually established, which includes a standard operating procedures of all aspects of the dispensing work. Supplemented by the pharmacist's regular professional training, and exchange of learning and quarterly performance appraisal incentive mechanism, the hospital pharmacy pharmacist prescription auditing capabilities was greatly improved, the occurrence of prescription dispensing errors was effective prevented.

Abstract:
Objective To investigate the role of clinical pharmacists in the treatment of severely infected patients. Method Clinical pharmacists took part in the consultations of empyrosis infected pediatric patient in department of burn, gave specific opinions on anti-infective drugs, and provided individualized dosage regimen for the pediatric patient. Results The pediatric patient was treated in time. Conclusion The participation of clinical pharmacists in clinical treatments could improve therapeutic level and promote rational drug use in hospitals.
Analysis of irrational prescriptions in emergency department

Abstract:
Objective To analyze the irrational drug use in the emergency department of our hospital, improve the quality of prescription and promote rational drug use in clinic. Methods 1 080 Prescriptions from January to June were randomly extracted from emergency department of our hospital for statistical analysis,and irrational prescriptions were reviewed. Results Of all the prescriptions reviewed, 4.35%(48/1 080) prescriptions were found to be irrational.The incidence of inappropriate dosage, contraindications and inappropriate drugs selection were high, which accounted for irrational prescription of 25.00%, 22.92%, 20.83%. Conclusions It was still some irrational drug use in emergency department. Therefore, prescription evaluation should be done to promote clinical rational use of drug, and guarantee patients with safety, reasonable and effective of drug use.

The experiment named anti-inflammatory effect of glucocorticoids was re-designed and implemented by undergraduate students. Abstract:
An attempt to apply designing experiment on determination two-component system by ultraviolet spectroscopy was made during the experiment teaching of analysis chemistry, whose purpose was to enable students to deepen understanding the principle in quantitative analysis of two-component system and enhance students' ability of analyzing and solving problems. The implementation methods and experience of the designing experiment were introduced in this paper and the improvement measures were discussed according to the students' feedback, which provided a basis for the designing experimentation course in teaching analysis chemistry.

Abstract:
Pharmacy undergraduate practice was important to improve the professional skills and research capacity of pharmacy undergraduates. Undergraduate practice could let the students practice and consolidate the basis knowledge of pharmacy which was learned in the class, learn and train the basic laboratory skills of pharmacy, the more important was to guide and establish a correct attitude of science research, cultivate the positivism of research and innovation ability, and improve the students' overall quality. According to some experiences in the work of undergraduates guidance, strengthening undergraduates' experimental operation skill, practicing the capability to exercise independent subject completion, advocating research and innovation capacity-building, and investigate the training mode in pharmacy undergraduates internship were discussed in this paper.
